I suspect the answer is no - the missing GTK dependencies probably cause an error immediately on load? The alternative would be supplying a script to launch Pinta that first checks for GTK somehow

There is a way for Windows to detect that some .Net is required to run an application, and prompts you to activate it before running it (I think it's for 4.0, since 4.5 is the default one in Win10? I don't remember).
(Of course, true, I don't remember if that happens on installation, like you resolve it now, or it can also happen when running the program itself)

Also, this happens even with GTK# installed.
I am triggering it by using Everything (voidtools.com) to search & execute it (instead of going there via explorer.exe or via a shortcut.lnk)
