Where is the dairy file located?

To use in an automatic backup program like Déjà Dup, so avoiding manually backups if it is possible.

Diary files can be located anywhere. You can move your existing diary anywhere you need to and open it with "Browse for a diary" option in the diary drop down menu.

This means that every time I press "Log off"(the little icon on the top right) automatically saved the new info on the file that I already save for example in the home folder?
Or I have to create a backup to save the new info?

Yes, when you log off or quit Lifeograph, it automatically saves your changes and also creates a backup of the diary in the same directory appending ".previousversion~" to its name. If you don't want to save a certain session, you should log out with "Log out without saving" option.
